Endocrine therapies bind directly to estrogen receptors on the surface of tumor cells. They do not affect the production of estrogen within the tumor cell itself. However, since the aromatase enzyme produces estradiol, it must be blocked to prevent excess estradiol from reaching the tumor cells.
